암세포 전이를 평가하는 방법-Invasion assay protocol-

Cancer의 가장 두려운 점은 바로 전이(metastasis)입니다. 전이과정이 일어나기 위해서는 암세포가 혈관벽을 뚫고 혈액으로 넘어가야하는데요. 이 과정을 Invasion이라고 합니다.

Invasion assay는 이 과정을 평가하는 방법으로 matrigel을 얼마나 파고 들어가 이동하는지 테스트 하는 실험입니다. 암세포를 연구하는데 기본적으로 실행되는 실험이기도 합니다.

 

 

1. matrigel coating 

-24well plate, pippet tip, 1.5ml 튜브, plate insert를 얼음에 박아둔다.

-matrigel은 하루전에 4도씨에서 녹인다.

-4도씨 RPMI를 준비한다.

-5mg/ml의 matrigel을 1mg/ml로 RPMI로 dilution (volumet 70ul) 한다. (70ul 기준- matrigel 18ul + 52ul media)

-조심스럽게 insert에 넣고 하루간 굳힌다.

 

 

 

2. Seeding 

-4x10^5 cell(A549 cells)을 각 insert에 넣고, 2일간 incubation한다.

-Lower chamber of the transwell is filled with 750ul of culture media containing 5ug/ml fibronectin, as adhesive subtrate.(serum포함)

(-fiblinectin stock 1mg/ml, final 5mg/ml : 3ul/600ul of media)

-Matrigel 을 pitting으로 제거하고 PBS로 두번 washing.

-Fix cell with 4% formaldehyde in PBS for 2 min (Both of upper and lower chamber).

-Remove formaldehyde and washing twice with PBS. 

-Permeabilize cell by 100% methanol for 20min at RT (Both of upper and lower chamber).

-Remove 100% methanol and washing twice with PBS.

-Add Giemsa stain 200-500ul for 15 min and washing twice with PBS. (호일로 빛 차단.)

-Scrape off non-invasive cell with cotton swabs ( 안쪽만 닦는다.)

 

-Count invasive cell under chamber.
